Biosynthesis occurs through salvage, Preiss-Handler, and de novo pathways. In mammals, the salvage pathway from nicotinamide predominates, and NAMPT is often described as rate-limiting. Nicotinamide riboside and nicotinic acid enter related routes that converge on NAD+ production. Tissue NAD+ concentrations vary widely and are maintained by a balance of synthesis and consumption. Some studies report age-related declines in certain tissues, but whether these changes cause disease or can be reversed to improve human health remains an open question.
NAD+ stands for nicotinamide adenine dinucleotide, the oxidized form of a coenzyme found in all living cells. The molecule consists of two nucleotides, adenine and nicotinamide ribose, joined through phosphate groups. Its chemical formula is C21H27N7O14P2, and the free acid has a molar mass near 663.43 grams per mole. In redox reactions, NAD+ accepts a hydride ion and becomes NADH. The pair NAD+ and NADH participates in hundreds of metabolic reactions, including steps in glycolysis, the citric acid cycle, and oxidative phosphorylation.
NAD+ also serves as a substrate for enzymes that cleave it, including sirtuins, PARPs, and CD38. These enzymes consume NAD+ and release nicotinamide and ADP-ribose or related products. The dual roles as redox cofactor and signaling substrate connect NAD+ to DNA repair, circadian regulation, and calcium signaling. Cellular NAD+ concentrations vary by tissue, time of day, and stress exposure. How these consumption pathways interact with redox balance remains an active area of research.
NAD+ is a dinucleotide composed of two nucleotides joined by a pyrophosphate linkage. One nucleotide contains adenine, and the other contains nicotinamide. The oxidized form carries a positive charge on the nicotinamide ring and is abbreviated NAD+. It functions as a cofactor in hydride-transfer reactions, accepting electrons in catabolic pathways. In cells, it interconverts with reduced NADH, forming a redox couple central to energy metabolism. The molecule is water-soluble and does not cross cell membranes freely without specific transport or precursor pathways.
The nicotinamide ring undergoes reversible reduction at the para position, converting NAD+ to NADH. This reaction transfers a hydride equivalent, not a free hydrogen atom or electron alone. Because the redox pair has a defined reduction potential, it links oxidation of fuels to respiratory chain activity. Many dehydrogenases use NAD+ as a co-substrate and produce NADH. The ratio of NAD+ to NADH reflects metabolic state and influences flux through several pathways.

Purified NAD+ is typically supplied as a white to off-white powder and stored desiccated at low temperature. Airtight containers limit moisture uptake, while protection from light reduces degradation of the nicotinamide ring. Aqueous stock solutions are less stable than solid material and are often aliquoted before freezing. Repeated freeze-thaw cycles can lower integrity, so working portions are kept separate. Purity is commonly checked by ultraviolet absorbance near 260 nm, high-performance liquid chromatography, or mass spectrometry.
Stability studies show that NAD+ can hydrolyze under prolonged heat, extreme pH, or microbial contamination. Phosphate buffers near neutral pH are often used for short-term handling, though exact stability depends on concentration, temperature, and matrix. In biological samples, endogenous enzymes can rapidly degrade NAD+, making cold chain and fast processing important. Analytical reports should state extraction conditions, internal standards, and validation parameters. Without those details, comparisons across studies remain difficult and potentially misleading.
Laboratory measurement of NAD+ often begins with rapid quenching of cell or tissue samples to prevent enzymatic conversion. Acidic or alkaline extraction can precipitate proteins, but the chosen method affects recovery of oxidized and reduced forms. Enzymatic cycling assays provide high sensitivity by amplifying a NAD+-dependent reaction. High-performance liquid chromatography and mass spectrometry offer separation and structural confirmation. Each method has trade-offs in throughput, specificity, and the ability to distinguish NAD+ from close analogues.
Quantification of NAD+ in biological samples usually relies on separation techniques coupled to sensitive detection. High-performance liquid chromatography with ultraviolet detection can measure the oxidized form by its absorbance near 260 nm, while mass spectrometry provides greater specificity and can distinguish NAD+ from close analogs. Enzymatic cycling assays use coupled dehydrogenase reactions to amplify signal and estimate NAD+ concentrations in cell or tissue extracts. Because NAD+ and NADH interconvert rapidly, sample preparation must quench metabolism quickly and preserve the redox state before analysis.
The stability of NAD+ depends on pH, temperature, light exposure, and the presence of degradative enzymes. Aqueous solutions are generally more stable under mildly acidic to neutral conditions and degrade faster under alkaline conditions or prolonged heat. The solid is hygroscopic and should be stored desiccated, often frozen, and protected from repeated freeze-thaw cycles. In laboratory handling, aliquots reduce repeated temperature changes, and chelating agents may limit metal-catalyzed hydrolysis in some buffers. These practices matter because even small amounts of NADH or hydrolysis products can interfere with quantitative assays.
Beyond redox catalysis, NAD+ is a substrate for enzymes that transfer ADP-ribose or remove acetyl groups from proteins. Sirtuins and poly(ADP-ribose) polymerases consume NAD+ and release nicotinamide as a byproduct. These reactions connect cellular energy status to gene regulation, DNA repair, and stress responses. Because NAD+ is used rather than merely recycled in such signaling, its concentration reflects both biosynthesis and consumption. The balance between salvage and de novo synthesis pathways determines available pools in different tissues.
Biosynthesis of NAD+ starts from nicotinamide, nicotinic acid, or nicotinamide riboside through salvage pathways. A rate-limiting enzyme, nicotinamide phosphoribosyltransferase, converts nicotinamide to nicotinamide mononucleotide. Further coupling with ATP yields NAD+. In mammals, the liver and muscle can synthesize NAD+ from dietary precursors, but tissue levels vary widely. Researchers study these pathways to understand age-related changes, metabolic disorders, and neurodegeneration. Direct causal links between NAD+ decline and disease remain an active area of investigation.
NAD+ is a dinucleotide composed of adenine, ribose, and nicotinamide groups joined by phosphate linkages. It serves as a coenzyme in oxidoreductase reactions, cycling between oxidized NAD+ and reduced NADH. The molecule is water-soluble and occurs in all living cells. Its nicotinamide ring accepts hydride ions during catabolic reactions, linking substrate oxidation to electron transport. This redox couple supports ATP production and helps maintain cytosolic and mitochondrial redox balance in many cell types.
Some opioid antagonists are not pure antagonists because they produce weak opioid partial agonist effects, and can produce analgesic effects when administered in high doses to individuals who have never taken opioids. Examples of such compounds include nalorphine and levallorphan. However, the analgesic effects from these specific drugs are limited and tend to be accompanied by dysphoria, most likely due to additional agonist action at the κ-opioid receptor. As they induce opioid withdrawal effects in people who are taking, or have recently used, opioid full agonists, these drugs are generally considered to be antagonists for practical purposes. The weak partial agonist effect can be useful for some purposes, and has previously been used for purposes such as long-term maintenance of former opioid addicts using nalorphine, however it can also have disadvantages such as worsening respiratory depression in patients who have overdosed on non-opioid sedatives such as alcohol or barbiturates. On the other hand, Naloxone has no partial agonist effects, and is in fact a partial inverse agonist at μ-opioid receptors, and so is the preferred antidote drug for treating opioid overdose. Naloxone and naltrexone are commonly used opioid antagonist drugs which are competitive antagonists that bind to the opioid receptors with higher affinity than agonists but do not activate the receptors. This effectively blocks the receptor, preventing the body from responding to opioids and endorphins. Naloxone is a drug used to treat opioid overdose.
Some foods, such as many cheeses, wines, and beers, are prepared by fermentation. This involves cultivating specific microorganisms to combat spoilage from other, less benign organisms. These microorganisms keep pathogens in check by producing acid or alcohol, which eventually creates an environment toxic for themselves and other microorganisms. Methods of fermentation include, but are not limited to, starter microorganisms, salt, hops, controlled (usually cool) temperatures and controlled (usually low) levels of oxygen. These methods are used to create the specific controlled conditions that will support the desirable organisms that produce food fit for human consumption. Fermentation is the microbial conversion of starch and sugars into alcohol. Not only can fermentation produce alcohol, but it can also be a valuable preservation technique. Fermentation can also make foods more nutritious and palatable. Water was also turned into alcoholic beverages through fermentation. When water is used to make beer, the boiling during the brewing process may kill bacteria that could make people sick. The barley and other ingredients also infuse the drink with nutrients, and the microorganisms can also produce vitamins as they ferment. However, the common belief that premodern people avoided drinking ordinary water is a myth. While people avoided drinking dirty or polluted water, they also avoided using it for the production of beer and wine. Water was visually inspected, smelled, tasted, filtered, and boiled if necessary.
=== Eligibility of minors === While the WPATH standards of care generally require the patient to have reached the age of medical majority, they include a separate section devoted to children and adolescents. Prepubescent children do not have access to medical intervention for gender-affirming therapy. After puberty, some medical intervention is available for adolescents depending on specific criteria for gender incongruence diagnosis, capacity for informed consent, and mental and physical health. According to a study by JAMA Pediatrics published in January 2025, less than 0.1% of adolescents covered by private medical insurance in the US take gender-affirming medication to treat gender dysphoria.
== Factor structure == Although Cohen et al. (1983) originally argued the PSS to be a unidimensional measure of perceived stress, the research community generally views the 14- and 10-item forms as two dimensional. The predominant forms consist of positively and negatively phrased items. Under exploratory factor analysis the negatively phrased items have been found to load onto a second factor separate from the positively phrased items, giving the appearance of a method effect. That is, a significant portion of the variability in the responses to a subset of the items is a product of how the items are phrased. However, inspection of the items suggests a substantive difference in the positively phrased items versus the negatively phrased items with the negatively phrased items on their face characterizing perceived helplessness while the positively phrased items may be characterizing perceived self-efficacy. Some support for this conceptualization has emerged. For example, Hewitt et al. (1992) found that the perceived helplessness items tended to predict depression in both men and women while the positively phrased items tended to predict depression in women only. The pattern of differential prediction of depression between genders tends to indicate the negatively phrased items are tapping something substantive rather than a method effect.
==== Water shortage and social unrest ==== Protests related to water shortages have become increasingly common in Iran. In July 2021, demonstrations erupted in Khuzestan when citizens accused the government of diverting water resources to support industries and other regions, leaving local communities without sufficient drinking water or irrigation supplies. Similar unrest has occurred in Sistan-Baluchestan, where residents face chronic water shortages exacerbated by mismanagement and illegal water extraction.

Trump emphasized that European nations should shoulder the primary burden of assistance and pressed Ukrainian president Volodymyr Zelenskyy to take greater responsibility in securing peace. He further hinted that Ukraine may eventually face difficult choices, including potential territorial concessions, to bring the conflict to an end. In September 2025, Trump urged Europe to stop buying Russian oil and start putting economic pressure on China for funding Russia's war effort. Treasury Secretary Scott Bessent said the Trump administration is "prepared to increase pressure on Russia, but we need our European partners to follow us". In September 2025, the Trump administration approved the delivery of the first two packages of weapons to Ukraine from US stockpiles totalling approximately $1 billion, which would be paid for by NATO allies, under the new mechanism called Prioritized Ukraine Requirements List (PURL). The mechanism aims to deliver aid worth up to $10 billion. On September 27, 2025, President Zelenskyy announced a $90 billion arms agreement with the United States in works. On October 22, 2025, the United States imposed sanctions on Russian energy companies Rosneft and Lukoil, affecting their customers in China and India. In November 2025, Trump proposed a peace plan that would require Ukraine to cede territory in the Donbas, including Ukrainian-controlled parts of Donetsk Oblast. The plan would also freeze the front lines in Kherson Oblast and Zaporizhzhia Oblast.
== In-utero development == Vernix is produced during a distinct phase of the epidermal development. Around the 21st week of gestation, periderm cells are being shed and replaced with stratum corneum; these shedding mix with secretions of sebum by the sebaceous glands to form vernix, which gradually covers the body in an anteroposterior and dorsoventral pattern. Vernix, in itself, is also believed to aid in the formation of stratum corneum. By early third trimester, the process is complete. Soon enough, part of the vernix is emulsified by increasing concentrations of pulmonary surfactants and desiccates, only to be consumed by the fetus; a corresponding increase in amniotic fluid turbidity is noticed.
==== Diverse food supply chain types ==== A mix of traditional, transitional and modern food supply chains can help buffer shocks and stresses of different types because the vulnerabilities and resilience capacities of food supply chains are shaped largely by their structural characteristics and product attributes:

=== Extracts === Products stating various E. longifolia extract ratios of 1:50, 1:100, and 1:200 are common on the market. However, extracts based on this ratio system are often misleading and hard to verify. Scientific research done on herbal products in general indicates that in many cases the content of bioactive constituents varies between products. Another option is for extraction techniques to use standardization methods to monitor the bioactive content and quality of the extract against standardization markers. Among standardization markers that have been used for E. longifolia are eurycomanone, total protein, total polysaccharide and glycosaponin, which have been recommended in a technical guideline developed by the Scientific and Industrial Research Institute of Malaysia.
== Signs and symptoms == The symptoms of ketoacidosis are variable depending on the underlying cause. The most common symptoms include nausea, vomiting, abdominal pain, and weakness. Breath may also develop the smell of acetone as it is a volatile ketone that can be exhaled. Rapid deep breathing, or Kussmaul breathing, may be present to compensate for the metabolic acidosis. Altered mental status is more common in diabetic than alcoholic ketoacidosis.
=== Oxytocin === Oxytocin is responsible for the milk let-down reflex as a response to neonate suckling. It is released from the posterior pituitary gland in a pulsatile manner, via stimulation of the vagus nerve. This causes myoepithelial cells, which surround the mammary alveoli, to contract. Oxytocin injections have been found to increase milk yield in cows. The role of oxytocin in the neonate is yet quite unclear, however we know that oxytocin has an important role in empathy and bonding between pairs.

Nicotinamide adenine dinucleotide, with the plus sign indicating the oxidized form. It is a coenzyme present in all living cells. The reduced form is NADH.
No. NAD+ is oxidized and accepts electrons, while NADH is reduced and carries them. Together they form a redox pair central to energy metabolism.
NAD+ itself is not a common dietary component in significant amounts. Precursors such as nicotinamide, nicotinic acid, and nicotinamide riboside can be converted through biosynthetic pathways. Direct absorption of intact NAD+ is limited.
NAD+ is the oxidized form, while NADH is the reduced form carrying an added hydride. The two form a redox pair that cells use in many energy-yielding reactions.
