Nicotinamide adenine dinucleotide, abbreviated NAD+, is a dinucleotide composed of two nucleotides joined by phosphate groups. One nucleotide contains adenine; the other contains nicotinamide. The molecule exists in oxidized (NAD+) and reduced (NADH) forms, and the reversible hydride transfer between them underlies many metabolic oxidation-reduction reactions. In cells, NAD+ serves as an electron acceptor in pathways such as glycolysis, the citric acid cycle, and oxidative phosphorylation. Its concentration and redox ratio vary by compartment, tissue, and metabolic state.
Beyond redox chemistry, NAD+ is consumed as a substrate by enzymes that transfer ADP-ribose or remove acetyl groups. Sirtuins use NAD+ in deacylation reactions, poly(ADP-ribose) polymerases use it in DNA damage responses, and CD38 hydrolases convert it to signaling metabolites. Because these enzymes compete for the same pool, changes in NAD+ availability can influence multiple cellular processes. The relative contribution of each consumption route differs by cell type and condition, and precise quantitative links remain an active area of study.
Research on NAD+ spans biochemistry, aging biology, and metabolism. Studies often examine how NAD+ levels change with age, diet, exercise, or disease states, and whether precursor supplementation alters those levels. Findings in animal models do not automatically translate to humans, and measurement methods vary across studies. Questions about tissue-specific effects, long-term consequences, and causal relationships remain open. NAD+ itself is not established as a single therapeutic agent with a broad clinical role.
The stability of NAD+ depends on pH, temperature, light exposure, and the presence of degradative enzymes. Aqueous solutions are generally more stable under mildly acidic to neutral conditions and degrade faster under alkaline conditions or prolonged heat. The solid is hygroscopic and should be stored desiccated, often frozen, and protected from repeated freeze-thaw cycles. In laboratory handling, aliquots reduce repeated temperature changes, and chelating agents may limit metal-catalyzed hydrolysis in some buffers. These practices matter because even small amounts of NADH or hydrolysis products can interfere with quantitative assays.
Quality control for NAD+ materials typically combines identity, purity, and water content checks. Identity may be confirmed by ultraviolet spectrum, retention time in chromatography, or mass accuracy, while purity is assessed by HPLC peak area or quantitative nuclear magnetic resonance. Residual water and solvents can affect molar calculations and enzyme assays, so Karl Fischer titration or thermogravimetric analysis may be used. Commercial materials vary in grade and counterion form, and published methods should specify the exact salt or hydrate when reporting concentrations. Regulatory status depends on intended use, with research reagents, dietary ingredients, and clinical products treated under different frameworks.

Beyond electron transfer, NAD+ serves as a substrate for enzymes that cleave it and attach its ADP-ribose portion to other molecules. This group includes poly(ADP-ribose) polymerases, CD38, and sirtuins. Such reactions consume NAD+ and can influence its availability for metabolism. Cells replenish NAD+ through a salvage pathway that recycles nicotinamide and through routes starting from tryptophan or vitamin B3 forms. How these synthesis and consumption routes are coordinated across tissues remains an active area of study, and compartment-specific concentrations are difficult to measure directly.
Nicotinamide adenine dinucleotide, abbreviated NAD+, is a dinucleotide built from adenine, nicotinamide, two ribose sugars, and two phosphate groups. The oxidized form carries a positive charge on the nicotinamide ring, while the reduced form, NADH, is neutral. This pair acts as a reversible electron carrier in cells. NAD+ is present in bacteria, plants, animals, and fungi. Its structure allows it to accept and donate electrons without being consumed in the reactions it supports.
In redox reactions, NAD+ accepts a hydride ion, which consists of two electrons and one proton. The hydride adds to the nicotinamide ring at a specific carbon, converting NAD+ into NADH. Dehydrogenase enzymes use this step in glycolysis, the citric acid cycle, and fatty acid oxidation. NADH later donates electrons to the mitochondrial electron transport chain, helping to drive ATP synthesis. The balance between NAD+ and NADH reflects the metabolic state of a cell, and shifts in that balance can alter how pathways operate.
NAD+ is relatively unstable in aqueous solution, especially at neutral or alkaline pH and at elevated temperatures. It is typically stored dry, protected from light and moisture, and kept cold or frozen for long-term use. Solutions are often prepared fresh or buffered to mildly acidic pH to slow hydrolysis. Repeated freeze-thaw cycles can reduce integrity. Laboratories may verify concentration using ultraviolet absorbance at 259 nm or by enzymatic assay. These handling practices are general laboratory conventions rather than universal rules.
Research on NAD+ often examines changes with age, diet, exercise, and disease states, but causal relationships are difficult to establish. Some studies measure NAD+ levels, while others assess enzyme activity or downstream markers. In the literature, terms such as "NAD+ decline" and "NAD+ boosting" appear in both scientific and commercial contexts, sometimes without precise definitions. Whether changes in measured NAD+ directly produce health effects remains an open question. Results from cells, animals, and humans cannot be assumed to translate directly.
Commercial NAD+ is supplied as a solid, often as the free acid or a salt, and purity is verified by chromatographic methods. Laboratories typically store it desiccated at minus 20 degrees Celsius or below. Working solutions are prepared fresh because even sterile aqueous solutions can lose activity over hours to days depending on pH and temperature. Documentation may include a certificate of analysis, an assay value, and a recommended retest date. Researchers should verify identity and purity when results depend on precise cofactor concentrations.
NAD+ is commonly measured by high-performance liquid chromatography with ultraviolet detection, often at 254 or 260 nm. Enzymatic cycling assays provide higher sensitivity by coupling NAD+ to a reporter reaction. Mass spectrometry can distinguish NAD+ from close analogues and confirm isotope labeling. Sample preparation usually involves rapid quenching of metabolism to prevent interconversion with NADH. Because NAD+ and NADH differ by one hydride, extraction conditions strongly affect the measured ratio.
== Possible scenarios == According to Kardashev, the most important parameters to define the existence of a civilization are three: the presence of very powerful energy sources, the use of non-standard technologies, and the transmission of significant amounts of information of various kinds through space.
== Screening == In the US, the US Preventive Services Task Force (USPSTF) publishes guidelines recommending preventive screening for certain types of common cancers and other diseases. As of March 2019, the USPSTF does not recommend screening for sarcoma, possibly because it is a very rare type of cancer (see Epidemiology below). The American Cancer Society (ACS) also publishes guidelines recommending preventive screening for certain types of common cancers. Like the USPSTF, as of March 2019 ACS does not recommend preventive screening for sarcoma. However, patients with some inherited conditions, such as neurofibromatosis, may benefit from screening for development of cancers from pre-existing benign tumors called neurofibromas.
== Distribution among species == Liver glucokinase occurs widely but not universally throughout vertebrate species. The gene structure and amino acid sequence are highly conserved among most mammals (e.g., rat and human glucokinase is more than 80% homologous). However, there are some unusual exceptions: For example, it has not been discovered in cats and bats, though some reptiles, birds, amphibians, and fish have it. Whether glucokinase occurs similarly in the pancreas and other organs has not yet been determined. It has been postulated that the presence of glucokinase in liver reflects the ease with which carbohydrates can be included in the animals' diets.
During the 19th century, the Liberal Party was broadly in favour of what would today be called classical liberalism, supporting laissez-faire economic policies such as free trade and minimal government interference in the economy (this doctrine was usually termed Gladstonian liberalism after the Victorian era Liberal Prime Minister William Gladstone). The Liberal Party favoured social reform, personal liberty, reducing the powers of the Crown and the Church of England (many of them were nonconformists) and an extension of the electoral franchise. Sir William Harcourt, a prominent Liberal politician in the Victorian era, said this about liberalism in 1872: If there be any party which is more pledged than another to resist a policy of restrictive legislation, having for its object social coercion, that party is the Liberal party. (Cheers.) But liberty does not consist in making others do what you think right, (Hear, hear.) The difference between a free Government and a Government which is not free is principally this—that a Government which is not free interferes with everything it can, and a free Government interferes with nothing except what it must. A despotic Government tries to make everybody do what it wishes; a Liberal Government tries, as far as the safety of society will permit, to allow everybody to do as he wishes. It has been the tradition of the Liberal party consistently to maintain the doctrine of individual liberty. It is because they have done so that England is the place where people can do more what they please than in any other country in the world.

The European authorities are the European Medicines Agency (EMEA), which is manly responsible for the scientific evaluation of medicines developed by pharmaceutical companies for use in the European Union, and enforcing the Registration, Evaluation, Authorization and Restriction of Chemicals (REACH) regulation. The U.S. Pharmacopeia codifies quality standards for Active Pharmaceutical Ingredients. As these standards are observed worldwide, they also contribute to the emergence of a uniform worldwide set-up of the top tier fine chemical companies. In terms of size, resources, and complexity of the chemical process technologies mastered, the fine chemical companies can be broadly divided into three segments, each of them accounting for approximately the same turnover of about $10 billion. The top tier, about twenty companies, has sales in excess of $250 million per year (see Table 3). Most are not pure players but divisions or business units of large, multinational companies. Their share varies between one percent or less for BASF and Pfizer, all the way to 100% for Cambrex, USA; Divi's Laboratories, India, and F.I.S., Italy. All have extensive resources in terms of chemists and other specialists, plants, process knowledge, backwards integration, international presence, etc.
== Contraindications == Tapentadol is contraindicated in people with epilepsy or who are otherwise prone to seizures. It raises intracranial pressure so should not be used in people with head injuries, brain tumors, or other conditions which increase intracranial pressure. It increases the risk of respiratory depression so should not be used in people with asthma. As with other μ-opioid agonists, tapentadol may cause spasms of the sphincter of Oddi, and is therefore discouraged for use in patients with biliary tract disease such as both acute and chronic pancreatitis. People who are rapid or ultra rapid metabolizers for the CYP2C9, CYP2C19, and CYP2D6 enzymes may not respond adequately to tapentadol therapy. Due to reduced clearance, tapentadol should be administered with caution to people with moderate liver disease and not at all in people with severe liver disease.
In June 2013, the leadership of the Chechen Republic officially recognized that up to 1,700 Chechen natives were fighting in the Middle East region. In the same year, according to information disseminated by the militants' Internet resources, as well as in the Russian media, Rustam Gelaev, the son of the famous Chechen commander Ruslan (Khamzat) Gelaev, was killed in Syria. Rustam died fighting on the side of the Syrian opposition.
=== Estrogen and menopause === The liver is responsive to estrogen. Animal models of estrogen deficiency accumulate liver fat as well as inflammation and fibrosis. The protective effect of estrogen is the usual explanation to why women have a much lower incidence of MASLD than men, before menopause. On the other hand, women with an earlier age at menopause are at increased risk for MASLD, and post-menopausal women with MASLD are at greater risk to develop severe fibrosis. Studies of various forms of menopausal hormone replacement therapy as potential treatment for MASLD, have yielded conflicting results.

=== Research === He started his research in the chemistry of natural products. At Washington State University he established early steps in the metabolism of d-neomethyl-α-D-glucoside in pipermint (Mentha piperita) rhizomes via in vivo studies. Bhushan developed a de novo method for direct resolution of certain racemates by liquid chromatography. Later, the approach was applied for direct enantioseparation of several active pharmaceutical ingredients (APIs). It is now an established approach in literature. 1994 onwards, the method was extended to such resolutions by ligand exchange principle. The method is of significant importance to pharmaceutical industry and analytical laboratories associated with regulatory agencies for determination and control of enantiomeric purity (and isolation of native enantiomers) of a variety of APIs since many of them are marketed and administered as racemic mixture while only one enantiomer is therapeutically useful. Bhushan supervised the Ph.D. theses of > 30 scholars and has published more than 270 research papers.

NAD+ is a coenzyme found in all living cells. It carries electrons in metabolic reactions and also serves as a substrate for enzymes involved in signaling and DNA repair. Its oxidized and reduced forms are central to energy metabolism.
NAD+ is the oxidized form and NADH is the reduced form. The pair accepts and donates electrons in redox reactions. Their ratio helps indicate the metabolic state of a cell or compartment.
No. Nicotinamide mononucleotide (NMN) and nicotinamide riboside (NR) are precursors that cells can convert into NAD+. They are distinct molecules with different absorption and metabolism profiles.
Common methods include LC-MS, HPLC with UV detection, and enzymatic cycling assays. Rapid quenching is needed because NAD+ and NADH interconvert. The chosen method should be validated for the sample matrix.
