NAD+ is the oxidized form of nicotinamide adenine dinucleotide, a coenzyme built from two nucleotides joined by a phosphate linkage. One nucleotide carries adenine, and the other carries nicotinamide; the plus sign denotes a formal positive charge on the nicotinamide ring, not a free proton. In cells, NAD+ and its reduced partner NADH form a reversible redox pair. That pair participates in electron transfer reactions throughout metabolism. The abbreviation NAD+ is common in biochemistry, while NAD(H) sometimes denotes the combined pool.
The molecule was first described in the early twentieth century as a factor that promoted fermentation in yeast extracts. Later work linked it to hydrogen transfer and to the oxidation of nutrients in living tissues. Its structure was resolved as a dinucleotide, which explained why it could accept and donate electrons at specific enzyme sites. Today, NAD+ is recognized as a central substrate and signaling precursor, not merely a metabolic cofactor. Whether all observed NAD+ changes reflect causal signaling remains an open question.
NAD+ stands for nicotinamide adenine dinucleotide, the oxidized form of a coenzyme found in all living cells. The molecule consists of two nucleotides, adenine and nicotinamide ribose, joined through phosphate groups. Its chemical formula is C21H27N7O14P2, and the free acid has a molar mass near 663.43 grams per mole. In redox reactions, NAD+ accepts a hydride ion and becomes NADH. The pair NAD+ and NADH participates in hundreds of metabolic reactions, including steps in glycolysis, the citric acid cycle, and oxidative phosphorylation.
In cells, NAD+ functions primarily as an electron carrier. Dehydrogenase enzymes in glycolysis and the citric acid cycle transfer hydride from substrates to NAD+, producing NADH. NADH then delivers electrons to the mitochondrial respiratory chain, supporting ATP synthesis. In fermentation, NADH is reoxidized to NAD+ so that glycolysis can continue. The balance between NAD+ and NADH helps set metabolic flux. Beyond redox, NAD+ serves as a substrate for enzymes that cleave it, including sirtuins, poly(ADP-ribose) polymerases, and CD38. These reactions consume NAD+ and release nicotinamide and ADP-ribose products.
| Property | Value | Notes |
|---|---|---|
| Chemical name | Nicotinamide adenine dinucleotide (oxidized form) | NAD+ denotes the oxidized redox state |
| Common synonyms | Diphosphopyridine nucleotide; coenzyme I | Older names appear in historical literature |
| Molar mass | About 663.43 g/mol | Free acid value; salts and hydrates differ |
| Appearance | White to off-white powder | The purified solid is white; solutions are clear |
| Solubility | Highly soluble in water | Aqueous buffers are common laboratory solvents |
Purified NAD+ is typically supplied as a white to off-white powder and stored desiccated at low temperature. Airtight containers limit moisture uptake, while protection from light reduces degradation of the nicotinamide ring. Aqueous stock solutions are less stable than solid material and are often aliquoted before freezing. Repeated freeze-thaw cycles can lower integrity, so working portions are kept separate. Purity is commonly checked by ultraviolet absorbance near 260 nm, high-performance liquid chromatography, or mass spectrometry.
Stability studies show that NAD+ can hydrolyze under prolonged heat, extreme pH, or microbial contamination. Phosphate buffers near neutral pH are often used for short-term handling, though exact stability depends on concentration, temperature, and matrix. In biological samples, endogenous enzymes can rapidly degrade NAD+, making cold chain and fast processing important. Analytical reports should state extraction conditions, internal standards, and validation parameters. Without those details, comparisons across studies remain difficult and potentially misleading.
Biosynthesis of NAD+ starts from nicotinamide, nicotinic acid, or nicotinamide riboside through salvage pathways. A rate-limiting enzyme, nicotinamide phosphoribosyltransferase, converts nicotinamide to nicotinamide mononucleotide. Further coupling with ATP yields NAD+. In mammals, the liver and muscle can synthesize NAD+ from dietary precursors, but tissue levels vary widely. Researchers study these pathways to understand age-related changes, metabolic disorders, and neurodegeneration. Direct causal links between NAD+ decline and disease remain an active area of investigation.
NAD+ is a dinucleotide composed of adenine, ribose, and nicotinamide groups joined by phosphate linkages. It serves as a coenzyme in oxidoreductase reactions, cycling between oxidized NAD+ and reduced NADH. The molecule is water-soluble and occurs in all living cells. Its nicotinamide ring accepts hydride ions during catabolic reactions, linking substrate oxidation to electron transport. This redox couple supports ATP production and helps maintain cytosolic and mitochondrial redox balance in many cell types.
Beyond redox catalysis, NAD+ is a substrate for enzymes that transfer ADP-ribose or remove acetyl groups from proteins. Sirtuins and poly(ADP-ribose) polymerases consume NAD+ and release nicotinamide as a byproduct. These reactions connect cellular energy status to gene regulation, DNA repair, and stress responses. Because NAD+ is used rather than merely recycled in such signaling, its concentration reflects both biosynthesis and consumption. The balance between salvage and de novo synthesis pathways determines available pools in different tissues.
== Diagnosis == Patient may present with symptomatic anemia or with sickle crises. In the United States and other countries with new-born screening programs, the disease may be identified in neonates. Diagnostic tests include DNA sequencing, hemoglobin electrophoresis, and high-performance liquid chromatography.
=== In pregnancy === For women who are pregnant and already have MG, in a third of cases, they have been known to experience an exacerbation of their symptoms, and in those cases, it usually occurs in the first trimester of pregnancy. Signs and symptoms in pregnant mothers tend to improve during the second and third trimesters. Complete remission can occur in some mothers. Immunosuppressive therapy should be maintained throughout pregnancy, as this reduces the chance of neonatal muscle weakness, and controls the mother's myasthenia. About 10–20% of infants with mothers affected by the condition are born with transient neonatal myasthenia gravis (TNMG), which generally produces feeding and respiratory difficulties that develop about 12 hours to several days after birth. A child with TNMG typically responds very well to acetylcholinesterase inhibitors, and the condition generally resolves over a period of three weeks, as the antibodies diminish, and generally does not result in any complications. However, a small percentage of fetuses and newborns with TNMG, particularly those who have antibodies directed against the fetal form of the AChR (their disorder is a subtype of TNMG termed the "acetylcholine receptor inactivation syndrome") have a more severe form of TNMG which includes weakness in skeletal muscles regulating breathing, respiratory failure, and various deformities such as arthrogryposis multiplex congenita. In some of these cases, the mother remains asymptomatic.

An isotope (nuclide) is specified by the name of the element (which implies the atomic number) followed by a hyphen and the mass number (e.g. helium-3, helium-4, carbon-12, carbon-14, uranium-235 and uranium-239). When a element symbol is used, e.g. "C" for carbon, it is standard to use "AZE notation" which has the form AZE, where A is the mass number written as a superscript, Z is the atomic number written as a subscript, and E is the element symbol (e.g. 32He, 42He, 126C, 146C, 23592U, and 23992U). Because the atomic number is already fixed by the element symbol, it is common to state only the mass number in the superscript and leave out the atomic number subscript (e.g. 3He, 4He, 12C, 14C, 235U, and 239U). The letter m (for metastable) is appended after the mass number to indicate a nuclear isomer, a metastable or energetically excited nuclear state (as opposed to the lowest-energy ground state), for example 180m73Ta (tantalum-180m); a number can be appended to it to distinguish different metastable states, though this is rare in practice. The common pronunciation of the AZE notation is different from how it is written: 42He is commonly pronounced helium-four instead of four-two-helium, and 23592U uranium two-thirty-five (American English) or uranium-two-three-five (British) instead of 235-92-uranium or 235-uranium. This is not an error but the original spoken usage for isotope names, originating before AZE notation became established.
Dimethocaine, also known as DMC or larocaine, is a compound with local anesthetic and stimulatory effects. The stimulatory effect resembles that of cocaine, although less potent. Similarly to cocaine, dimethocaine is addictive due to its stimulation of the reward pathway in the brain. However, dimethocaine is a legal cocaine replacement in some countries and is listed by the European Monitoring Centre for Drugs and Drug Addiction (EMCDDA) under the "synthetic cocaine derivatives" category. The structure of dimethocaine, being a 4-aminobenzoic acid ester, resembles that of procaine. It is found as a white powder at room temperature. When a product sold online in the UK in June 2010, advertised as dimethocaine was tested, it was found to be a mixture of caffeine and lidocaine, and the lack of any dopaminergic stimulant ingredient in such mixes may explain the limited recreational effects reported by many users. Other samples tested have however been shown to contain genuine dimethocaine, and one branded "bath salt" product containing primarily dimethocaine as the active ingredient, was noted to have been particularly subject to abuse by intravenous drug users in Ireland.

=== Processing === Water is removed from the sample in successive stages by the use of increasing concentrations of alcohol. Xylene is used in the last dehydration phase instead of alcohol - this is because the wax used in the next stage is soluble in xylene where it is not in alcohol allowing wax to permeate (infiltrate) the specimen. This process is generally automated and done overnight. The wax infiltrated specimen is then transferred to an individual specimen embedding (usually metal) container. Finally, molten wax is introduced around the specimen in the container and cooled to solidification so as to embed it in the wax block. This process is needed to provide a properly oriented sample sturdy enough for obtaining a thin microtome section(s) for the slide. Once the wax embedded block is finished, sections will be cut from it and usually placed to float on a water bath surface which spreads the section out. This is usually done by hand and is a skilled job (histotechnologist) with the lab personnel making choices about which parts of the specimen microtome wax ribbon to place on slides. A number of slides will usually be prepared from different levels throughout the block. After this the thin section mounted slide is stained and a protective cover slip is mounted on it. For common stains, an automatic process is normally used; but rarely used stains are often done by hand.

==== Caribbean and Latin America ==== Cream soda is usually served as a "red pop", particularly Fanta's Red Cream Soda. Champagne cola (also spelled "kola"), a soft drink similar to cream soda, is ubiquitous across the region. In the Caribbean there are several popular brands of clear, vanilla-flavored cream soda.
Structurally, the conjugated proteins are important in providing stability to cellular components and help intercellular communication which is why glycoproteins are commonly found in cell membranes and extracellular matrices.
Franz Joseph was born on 18 August 1830 in the Schönbrunn Palace in Vienna (on the 65th anniversary of the death of Francis of Lorraine) as the eldest son of Archduke Franz Karl (the younger son of Francis I), and Sophie, Princess of Bavaria. Because his uncle, reigning from 1835 as the Emperor Ferdinand, was disabled by seizures, and his father unambitious and retiring, the mother of the young Archduke "Franzi" brought him up as a future emperor, with emphasis on devotion, responsibility and diligence. For this reason, Franz Joseph was consistently built up as a potential successor to the imperial throne by his politically ambitious mother from early childhood. Up to the age of 7, little "Franzi" was brought up in the care of the nanny ("Aja") Louise von Sturmfeder. Then the "state education" began, the central contents of which were "sense of duty", religiosity and dynastic awareness. The theologian Joseph Othmar von Rauscher conveyed to him the inviolable understanding of rulership of divine origin (divine grace), and therefore a belief that no participation of the population in rulership in the form of parliaments was required. The educators Heinrich Franz von Bombelles and Colonel Johann Baptist Coronini-Cronberg ordered Archduke Franz to study an enormous amount of time, which initially comprised 18 hours per week and was expanded to 50 hours per week by the age of 16.
=== Protein composition === EPCIP in humans has higher cysteine and lower valine concentrations than expected compared to other human proteins. This trend, as showed in Table 1, is the same for other mammals. It does not, however, occur in taxa other than mammalia.
=== European Union === In 2000, the European Union (EU) enacted similar legislation, Regulation(EC) No 141/2000, which refers to drugs developed to treat rare diseases to as "orphan medicinal products". The EU's definition of an orphan condition is broader than that of the US, in that it also covers some tropical diseases that are primarily found in developing nations. Orphan drug status granted by the European Commission gives marketing exclusivity in the EU for 10 years after approval. The EU's legislation is administered by the Committee on Orphan Medicinal Products of the European Medicines Agency (EMA). In late 2007 the FDA and EMA agreed to use a common application process for both agencies to make it easier for manufacturers to apply for orphan drug status but, while continuing two separate approval processes.
