==== Premature aging ==== Premature aging syndromes including Werner syndrome, Progeria, Ataxia telangiectasia, Ataxia-telangiectasia like disorder, Bloom syndrome, Fanconi anemia and Nijmegen breakage syndrome are associated with short telomeres. However, the genes that have mutated in these diseases all have roles in the repair of DNA damage and the increased DNA damage may, itself, be a factor in the premature aging (see DNA damage theory of aging). An additional role in maintaining telomere length is an active area of investigation.

A study treated non-diabetic, healthy men with the GLP-1 receptor antagonist (i.e., blocker of receptor activation) exendin(9-39)NH2a (also termed avexitide), the GIP receptor antagonist GIP(3-30)NH2, or both antagonists and challenged them with an oral glucose tolerance test. Men treated with either agent responded to the tolerance test with modest decreases in blood insulin levels and modest increases in blood glucose levels. However, men treated with both antagonists responded with very low insulin and very high glucose blood levels: their responses were similar to those in individuals with type 2 diabetes. This study shows that 1) the stimulation of the FFAR2 on K and L cells by SCFAs underlies the differences between oral and intravenous glucose challenges defined by the incretin effect and 2) FFAR2 functions to regulate blood insulin and glucose levels. This does not prove that type 2 diabetes is a FFAR2-incretin disease: post-feeding secretion of the incretins (i.e., GLP-1 and GIP) is impaired in type 2 diabetes, but the impairment appears to result primarily from decreases in the responsiveness of pancreas alpha cells to GLP-1. This conclusion is supported by studies showing that type 2 diabetic individuals who are treated with large amounts of GLP-1 and challenged with intravenous glucose show changes in blood insulin and glucose levels that are similar to those in non-diabetic individuals. Indeed, GLP-1 agonists, e.g., Dulaglutide, and a first-in-kind GLP-1 and GIP agonist, Tirzepatide, are used to treat type 2 diabetes.

Promotion of the Mediterranean diets among the obese may lower the risk of heart disease. Decreased intake of sweet drinks is also related to weight-loss. Success rates of long-term weight loss maintenance with lifestyle changes are low, ranging from 2–20%. Dietary and lifestyle changes are effective in limiting excessive weight gain in pregnancy and improve outcomes for both the mother and the child. Intensive behavioral counseling is recommended in those who are both obese and have other risk factors for heart disease.
Severe amoebic colitis: Fulminant amoebic colitis is associated with high case fatality and can occur in patients infected with the parasite Entamoeba histolytica after exposure to corticosteroid medications. Neuropsychiatric: steroid psychosis, and anxiety, depression. Therapeutic doses may cause a feeling of artificial well-being known as "steroid euphoria". Rare case reports of corticosteroid misuse, addiction, and dependence due to euphoria exist. The neuropsychiatric effects are partly mediated by sensitization of the body to the actions of adrenaline. Therapeutically, the bulk of corticosteroid dose is given in the morning to mimic the body's diurnal rhythm; if given at night, the feeling of being energized will interfere with sleep. Cardiovascular: Corticosteroids can cause sodium retention through a direct action on the kidney, in a manner analogous to the mineralocorticoid aldosterone. This can result in fluid retention and hypertension. Metabolic: Corticosteroids cause a movement of body fat to the face and torso, resulting in "moon face", "buffalo hump", and "pot belly" or "beer belly", and cause movement of body fat away from the limbs. This has been termed corticosteroid-induced lipodystrophy. Due to the diversion of amino-acids to glucose, they are considered anti-anabolic, and long term therapy can cause muscle wasting (muscle atrophy).
=== Pulp stones === Pulp stones are calcified masses that occur in the pulp, either in the apical or coronal portions. They are classified according to their structure or location. According to their location, pulp stones can be classed either as free (completely surrounded by pulp), embedded (surrounded by dentine tissue) or adherent (attached to pulp wall continuous with dentine, but not fully enclosed). Depending on the structure, they are either true (dentine lined by odontoblasts), false (formed from degenerating cells that mineralise) or diffuse (more irregular in shape to false stones). In late 1991, Fripp asked former Japan singer David Sylvian to join a new King Crimson line-up. Sylvian declined the offer, but agreed to work with Fripp on a collaborative album; the resulting album The First Day was issued in 1993 under the name Sylvian/Fripp. In June 1993, Fripp began to assemble a larger version of King Crimson featuring Belew and Levin, Chapman Stick player Trey Gunn (a veteran of Fripp's Guitar Craft courses and a session musician on The First Day) and drummer Jerry Marotta, with whom Fripp and Levin had played in Peter Gabriel's band. After Sylvian/Fripp's closing concerts at the Royal Albert Hall in December 1993, Fripp decided to ask the tour's drummer Pat Mastelotto, formerly of Mr. Mister, to join instead of Marotta. Bruford would become the last member of the 1980s quartet to return to the band. Bruford later said he had lobbied Fripp at the last minute because he believed that Crimson was still very much "his gig", and that Fripp had come up with a philosophical explanation for utilizing both Mastelotto and Bruford later. One of the conditions Fripp imposed upon Bruford if he were to return was to give up all creative control to Fripp. Fripp later explained that he had a vision of a "Double Trio" with two guitarists, two bassists, and two drummers while driving along the Chalke Valley in Wiltshire one afternoon in 1992. === Cardiovascular health === Matrix Gla protein is a vitamin K-dependent protein found in bone, but also in soft tissues such as arteries, where it appears to function as an anti-calcification protein. In animal studies, animals that lack the gene for MGP exhibit calcification of arteries and other soft tissues. In humans, Keutel syndrome is a rare recessive genetic disorder associated with abnormalities in the gene coding for MGP and characterized by abnormal diffuse cartilage calcification. These observations led to a theory that in humans, inadequately carboxylated MGP, due to low dietary intake of the vitamin, could result in increased risk of arterial calcification and coronary heart disease. In meta-analyses of population studies, low intake of vitamin K was associated with inactive MGP, arterial calcification and arterial stiffness. Lower dietary intakes of vitamin K1 and vitamin K2 were also associated with higher coronary heart disease. When blood concentration of circulating vitamin K1 was assessed there was an increased risk in all cause mortality linked to low concentration. In contrast to these population studies, a review of randomized trials using supplementation with either vitamin K1 or vitamin K2 reported no role in mitigating vascular calcification or reducing arterial stiffness. The trials were too short to assess any impact on coronary heart disease or mortality.

=== Trophoblast differentiation === In addition, HAND1 is uniquely expressed in trophoblasts and is essential for early trophoblast giant cell differentiation. Trophoblast giant cells are necessary in order for placental development to proceed, participating in vital processes such as blastocyst implantation, remodeling of the maternal decidua, and secretion of hormones. The importance of this relationship is demonstrated in HAND1-null mutant mice, which display significant abnormalities in trophoblast development, such as a reduced ectoplacental cone, thin parietal yolk sac, and reduced density of trophoblast giant cells. These homozygous HAND1-null mutant embryos were arrested by E7.5 of gestation, though could be saved by contribution of wild-type cells to the trophoblast.
