Measuring NAD+ in biological samples requires rapid processing because the compound can degrade or interconvert after collection. Common approaches include enzymatic cycling assays, high-performance liquid chromatography, and mass spectrometry. Each method has different sensitivity, specificity, and susceptibility to interference from related nucleotides. Sample type matters: cultured cells, animal tissues, and human blood present distinct challenges. Reported values can vary widely across laboratories because of differences in extraction, normalization, and analytical platform. Standardization remains an open issue in the field.
NAD+ is relatively unstable in aqueous solution, especially at neutral or alkaline pH and at elevated temperatures. It is typically stored dry, protected from light and moisture, and kept cold or frozen for long-term use. Solutions are often prepared fresh or buffered to mildly acidic pH to slow hydrolysis. Repeated freeze-thaw cycles can reduce integrity. Laboratories may verify concentration using ultraviolet absorbance at 259 nm or by enzymatic assay. These handling practices are general laboratory conventions rather than universal rules.
Research on NAD+ often examines changes with age, diet, exercise, and disease states, but causal relationships are difficult to establish. Some studies measure NAD+ levels, while others assess enzyme activity or downstream markers. In the literature, terms such as "NAD+ decline" and "NAD+ boosting" appear in both scientific and commercial contexts, sometimes without precise definitions. Whether changes in measured NAD+ directly produce health effects remains an open question. Results from cells, animals, and humans cannot be assumed to translate directly.
Laboratory measurement of NAD+ often begins with rapid quenching of cell or tissue samples to prevent enzymatic conversion. Acidic or alkaline extraction can precipitate proteins, but the chosen method affects recovery of oxidized and reduced forms. Enzymatic cycling assays provide high sensitivity by amplifying a NAD+-dependent reaction. High-performance liquid chromatography and mass spectrometry offer separation and structural confirmation. Each method has trade-offs in throughput, specificity, and the ability to distinguish NAD+ from close analogues.
Purified NAD+ is typically supplied as a white to off-white powder and stored desiccated at low temperature. Airtight containers limit moisture uptake, while protection from light reduces degradation of the nicotinamide ring. Aqueous stock solutions are less stable than solid material and are often aliquoted before freezing. Repeated freeze-thaw cycles can lower integrity, so working portions are kept separate. Purity is commonly checked by ultraviolet absorbance near 260 nm, high-performance liquid chromatography, or mass spectrometry.
Stability studies show that NAD+ can hydrolyze under prolonged heat, extreme pH, or microbial contamination. Phosphate buffers near neutral pH are often used for short-term handling, though exact stability depends on concentration, temperature, and matrix. In biological samples, endogenous enzymes can rapidly degrade NAD+, making cold chain and fast processing important. Analytical reports should state extraction conditions, internal standards, and validation parameters. Without those details, comparisons across studies remain difficult and potentially misleading.

The molecule was first described in the early twentieth century as a factor that promoted fermentation in yeast extracts. Later work linked it to hydrogen transfer and to the oxidation of nutrients in living tissues. Its structure was resolved as a dinucleotide, which explained why it could accept and donate electrons at specific enzyme sites. Today, NAD+ is recognized as a central substrate and signaling precursor, not merely a metabolic cofactor. Whether all observed NAD+ changes reflect causal signaling remains an open question.
Related compounds include NADH, the reduced form, and NADP+, which carries an additional phosphate group. NADP+ and NADPH often serve in biosynthetic and antioxidant reactions, while NAD+ and NADH are more associated with energy-yielding catabolism. Nicotinamide, nicotinic acid, and nicotinamide riboside are precursors that can enter salvage pathways. The exact contribution of dietary precursors to tissue NAD+ pools is an area of active investigation. Some studies measure labeled precursors to trace those routes.
Laboratory measurement of NAD+ usually begins with rapid sample quenching because the molecule can change form after collection. Enzymatic cycling assays amplify signal through coupled reactions and are suited to small samples. High-performance liquid chromatography with ultraviolet detection separates NAD+ from related nucleotides. Liquid chromatography-mass spectrometry offers higher specificity and can distinguish NAD+ from close analogs. Each method has trade-offs in sensitivity, throughput, and equipment needs, so reported values depend heavily on extraction and detection choices.
Stability of NAD+ depends on form, temperature, pH, and water content. The solid is generally more stable than solutions, and it should be kept dry and cold. In solution, hydrolysis can cleave the dinucleotide, especially under alkaline conditions or at elevated temperature. Light exposure may also contribute to degradation. Buffers, chelating agents, and sterile handling can reduce losses, but no single condition preserves all preparations indefinitely. Researchers often prepare working solutions shortly before use and verify activity or purity after storage.
Quality control for NAD+ relies on identity, purity, and functional tests. A certificate of analysis may report high-performance liquid chromatography purity, ultraviolet spectrum, water content, and residual solvents. Because NAD+ is hygroscopic, gravimetric values can shift as material absorbs water, so purity should be interpreted alongside storage history. Mass spectrometry confirms molecular identity, while enzymatic assays show whether the material supports dehydrogenase activity. Commercial material is available as the free acid and as salts, and the counterion affects molecular weight, solubility, and how concentrations are calculated.
Lorazepam serum levels are proportional to the dose administered. Giving 2 mg oral lorazepam will result in a peak total serum level of around 20 ng/mL around two hours later, half of which is lorazepam, half its inactive metabolite, lorazepam-glucuronide. A similar lorazepam dose given intravenously will result in an earlier and higher peak serum level, with a higher relative proportion of unmetabolised (active) lorazepam. On regular administration, maximum serum levels are attained after three days. Longer-term use, up to six months, does not result in further accumulation. On discontinuation, lorazepam serum levels become negligible after three days and undetectable after about a week. Lorazepam is metabolized in the liver by conjugation into inactive lorazepam-glucuronide. This metabolism does not involve liver oxidation, so is relatively unaffected by reduced liver function. Lorazepam-glucuronide is more water-soluble than its precursor, so gets more widely distributed in the body, leading to a longer half-life than lorazepam. Lorazepam-glucuronide is eventually excreted by the kidneys, and, because of its tissue accumulation, it remains detectable, particularly in the urine, for substantially longer than lorazepam.
In 1998, at the 5th Conference of the Inter‑American Masonic Confederation (CMI) (Spanish: Confederación Masónica Interamericana), the 95 member bodies of CMI recognized the Grand Lodge of Cuba as "regular and correct," and rejected the regularity claims of the Grand Lodge of Cuba in Exile. In 2000, the Cuban government pressured the Grand Lodge of Cuba to elect a man named José Manuel Collera Venta as Grand Master. In 2005, when the Grand Lodge of Cuba attempted to expel Venta from Freemasonry "for an undisclosed reason," the Cuban government again intervened and stopped that from happening. By May 8, 2000, two factions existed within Cuban Freemasonry. One faction based in the National Temple headquarters preferred not to display any dissent against the government for the fear that their Lodges would be shuttered, but another faction within the leadership of the local Lodges advocated that Freemasons should be faithful to their statues, maintaining an impartial political position, no matter the consequences that might befall them. Lodges further away from Havana had more freedom due to less infiltration by government agents. In 2002, the Grand Lodge of Cuba reestablished its relationship with the Grand Lodge of Italy and the Grand Orient of Italy. By the year 2008, there were 29,110 Freemasons in Cuba, compared to 1980, when there were only 19,690.
Wounded healer is a term created by psychiatrist Carl Jung. The idea states that an analyst is compelled to treat patients because the analyst himself is "wounded." The idea may have Greek mythology origins. Victor et al. (2022) found that 82% of applied psychology graduate students and faculty members in the United States and Canada experienced mental health conditions at some point in their lives. As an example, of the "wounded healer phenomenon" between an analyst and their analyzed:

=== China === The People's Republic of China defines a VOC as those compounds that have "originated from automobiles, industrial production and civilian use, burning of all types of fuels, storage and transportation of oils, fitment finish, coating for furniture and machines, cooking oil fume and fine particles (PM 2.5)", and similar sources. In July 2018, the State Council released the "Three-Year Action Plan for Winning the Blue Sky Defence War", planning to reduce 2015 VOC emissions 10% by 2020.
== Regulation == Allosteric regulation by metabolites. The regulation of the citric acid cycle is largely determined by product inhibition and substrate availability. If the cycle were permitted to run unchecked, large amounts of metabolic energy could be wasted in overproduction of reduced coenzyme such as NADH and ATP. The major eventual substrate of the cycle is ADP which gets converted to ATP. A reduced amount of ADP causes accumulation of precursor NADH which in turn can inhibit a number of enzymes. NADH, a product of all dehydrogenases in the citric acid cycle with the exception of succinate dehydrogenase, inhibits pyruvate dehydrogenase, isocitrate dehydrogenase, α-ketoglutarate dehydrogenase, and also citrate synthase. Acetyl-coA inhibits pyruvate dehydrogenase, while succinyl-CoA inhibits alpha-ketoglutarate dehydrogenase and citrate synthase. When tested in vitro with TCA enzymes, ATP inhibits citrate synthase and α-ketoglutarate dehydrogenase; however, ATP levels do not change more than 10% in vivo between rest and vigorous exercise. There is no known allosteric mechanism that can account for large changes in reaction rate from an allosteric effector whose concentration changes less than 10%. Citrate is used for feedback inhibition, as it inhibits phosphofructokinase, an enzyme involved in glycolysis that catalyses formation of fructose 1,6-bisphosphate, a precursor of pyruvate. This prevents a constant high rate of flux when there is an accumulation of citrate and a decrease in substrate for the enzyme. Regulation by calcium.
According to oil workers in the area, Iraq's slant drilling claim was fabricated, as "oil flows easily from the Rumaila field without any need for these techniques." At the same time, Saddam looked for closer ties with those Arab states that had supported Iraq in the war. This move was supported by the US, who believed that Iraqi ties with pro-Western Gulf states would help bring and maintain Iraq inside the US' sphere of influence. In 1989, it appeared that Saudi–Iraqi relations, strong during the war, would be maintained. A pact of non-interference and non-aggression was signed between the countries, followed by a Kuwaiti-Iraqi deal for Iraq to supply Kuwait with water for drinking and irrigation, although a request for Kuwait to lease Iraq Umm Qasr was rejected. Saudi-backed development projects were hampered by Iraq's large debts, even with the demobilization of 200,000 soldiers. Iraq also looked to increase arms production so as to become an exporter, although the success of these projects was also restrained by Iraq's obligations; in Iraq, resentment to OPEC's controls mounted. Iraq's relations with its Arab neighbors, particularly Egypt, were degraded by mounting violence in Iraq against expatriate groups, who were well-employed during the war, by unemployed Iraqis, among them demobilized soldiers. These events drew little notice outside the Arab world because of fast-moving events directly related to the fall of Communism in Eastern Europe. However, the US did begin to condemn Iraq's human rights record, including the well-known use of torture.

In 2012, 37.1% of infants under six months of age were breastfeeding exclusively, which rose to 48% in 2022. The Sustainable Development Goals target is to achieve 70% by 2030. In the United States, the rate of women beginning to breastfeed was 76% in 2009, increasing to 83% in 2015, with 58% still breastfeeding at 6 months, although only 25% were still breastfeeding exclusively. African-American women have persistently low rates of breastfeeding compared to White and Hispanic American women. In 2014, 58.1% of African-American women breastfeed in the early postpartum period, compared to 77.7% of White women and 80.6% of Hispanic women. In 2019, 84.1% of U.S women giving birth initiated breastfeeding, with 87.4%, 85.5%, 73.6%, 90.3% and 83.1% of Hispanic, White, African-American, Asian and Multiracial mothers initiating, respectively. Rates of initiation among African-American mothers varied widely by state, with lows under 53% and highs over 90%. Breastfeeding rates in different parts of China vary considerably. Rates in the United Kingdom were the lowest in the world in 2015, with only 0.5% of mothers still breastfeeding at a year, while in Germany, 23% are doing so, 56% in Brazil, and 99% in Senegal. In Australia, for children born in 2004, more than 90% were initially breastfed. In Canada, for children born in 2005–06, more than 50% were only breastfed, and more than 15% received both breastmilk and other liquids by the age of 3 months.
During October and November 2005, Wenger became embroiled in a war of words with Chelsea manager José Mourinho. Mourinho accused Wenger of having an "unprofessional obsession" with Chelsea and labelled him a "voyeur". Mourinho was quoted as saying, "He's worried about us, he's always talking about us – it's [always] Chelsea". Wenger responded by pointing out he was only answering journalists' questions about Chelsea, and described Mourinho's attitude as "disrespectful". Mourinho later apologised and clarified that he regretted his "voyeur" comment; Wenger accepted the apology. In 2014, Mourinho reopened his feud with Wenger by calling him a "specialist in failure". This was in response to Wenger's comments that Chelsea were favourites for the Premier League and managers did not want to take responsibility if they failed to win the title. Wenger said Mourinho's comments were "silly and disrespectful" and had embarrassed Chelsea. When asked if he regretted his remark, Mourinho replied: "You have to ask him if he regrets the comments he made". In October 2014, during a Premier League match between Arsenal and Chelsea, Wenger was involved in a touchline spat with Mourinho. Wenger later apologised for his behaviour. Wenger has directed his anger towards referees when decisions have not gone his team's way. In August 2000, he was charged with "alleged threatening behaviour and physical intimidation" towards fourth official Paul Taylor, after Arsenal's 1–0 defeat at Sunderland on the opening day of the 2000–01 season.
==== 10 April ==== At midnight (local time), Hezbollah launched a rocket attack on the border town of Metula. Later on, sirens were heard in Tel Aviv, Haifa, and Ashdod in response to an incoming Hezbollah strike. Trump accused Iran of doing a "very poor job" in its handling of oil passing through the pivotal Strait of Hormuz, arguing it was "not the agreement we have". By 10 March 2026 only 15 ships made it through the Strait of Hormuz.

Researchers often use enzymatic cycling assays, liquid chromatography, or mass spectrometry. The choice depends on sample size, sensitivity needs, and available equipment. Because NAD+ can degrade quickly, rapid extraction and careful handling are important.
Differences can arise from sample type, extraction method, normalization strategy, and analytical platform. Time of day, diet, and physiological state may also matter. These factors make direct comparisons across studies difficult.
NAD+ is generally more stable when stored dry and cold, and it can degrade in aqueous solutions over time. Heat, light, and alkaline conditions can accelerate loss. Laboratory protocols therefore often recommend frozen storage and protection from light.
Many enzymes consume or produce NAD+ within seconds after a sample is collected. Quenching stops those reactions and helps preserve the ratio between oxidized and reduced forms. The exact quenching method depends on the tissue or cell type and the analytes of interest.
